目的 :U5 0 488H是阿片肽受体 κ亚型的选择性激动剂 ,本实验研究 U5 0 488H预处理是否可增强心肌细胞对热应激的耐受性 ,同时观察与热休克蛋白 70及 90表达的关系。方法 :采用分离的大鼠心肌细胞作为研究对象 ,以心肌细胞释放乳酸脱氢酶 (L DH)的多少反映其损伤程度并用 Western blot分析热休克蛋白表达的变化。结果 :在 5~ 30 μmol/ L 浓度范围内 ,U5 0 488H预处理 30 min剂量依赖性的减低心肌细胞热应激后 L DH的释放。同时热休克蛋白 70和 90的表达明显增加。结论 :U5 0 488H激活阿片受体后具有保护心肌细胞的作用 ,其保护机制可能与调节热休克蛋白的表达有关。
OBJECTIVE: U5 0 488H is a selective agonist of opioid receptor κ subtype. In this study, we investigated whether U5 0 488H pretreatment can enhance the tolerance of cardiomyocytes to heat stress and observe the effects of heat shock protein 70 and 90 The relationship between expression. Methods: Isolated rat cardiomyocytes were used as research objects. L DH was released from cardiomyocytes to reflect the degree of injury and the expression of heat shock protein was analyzed by Western blot. RESULTS: Pretreatment with U5 0 488H for 30 min reduced the L DH release in cardiomyocytes in a dose-dependent manner in the concentration range of 5 ~ 30 μmol / L. At the same time, the expression of heat shock proteins 70 and 90 increased significantly. CONCLUSION: U5 0 488H can activate cardiomyocytes after activation of opioid receptor, and its protective mechanism may be related to the regulation of heat shock protein expression.
