目的观察凋亡抑制基因生存素(Survivin)和E-钙粘附素(E-cadherin)在子宫腺肌病的表达及其相关性。方法以兰州市妇幼保健院2013年5月至2014年3月68例子宫病变患者为研究对象应用免疫组化En Vision二步法,检测38例子宫腺肌病病灶及在位内膜组织中Survivin及E-cadherin蛋白的表达,并与30例无症状肌瘤患者的正常子宫内膜组织进行对照;应用Western blot法检测Survivin及E-cadherin在子宫腺肌病病灶与正常内膜的表达。统计学分析以α=0.05为检验水准,当采用R×C表χ2检验的分割法时,检验水准调整为α’=0.0125。结果子宫腺肌病病灶组织中,Survivin蛋白的阳性表达率(89.47%)明显高于在位内膜(55.26%)及正常内膜(6.67%,P均<0.0125)。腺肌病病灶组织中E-cadherin蛋白阳性表达率(47.37%)明显低于在位内膜(71.05%)及正常内膜(96.67%,P均<0.0125)。腺肌病病灶中Survivin和E-cadherin表达呈线性负相关(r=-0.988,P<0.01);Western blot结果显示Survivin蛋白在腺肌病病灶的相对表达量明显高于正常内膜,E-cadherin在腺肌病病灶的相对表达量明显低于正常内膜。结论 Survivin及E-cadherin可能与子宫腺肌病的发生、发展有关。
Objective To observe the expression of survivin and E-cadherin in adenomyosis and its relationship. Methods 68 patients with uterine lesions from May 2013 to March 2014 in Lanzhou Maternal and Child Health Hospital were enrolled in this study. Immunohistochemistry En Vision immunohistochemistry was used to detect the expression of Survivin in 38 cases of adenomyosis and eutopic endometrium And E-cadherin protein expression were detected and compared with normal endometrium in 30 patients with asymptomatic fibroids. The expression of Survivin and E-cadherin in adenomyosis and normal endometrium was detected by Western blot. Statistical analysis with α = 0.05 as the test level, when using R × C table χ2 test segmentation method, the test level adjusted to α ’= 0.0125. Results The positive rate of Survivin protein expression in adenomyosis was 89.47%, which was significantly higher than that in eutopic endometrium (55.26%) and normal endometrium (6.67%, P <0.0125). The positive expression rate of E-cadherin protein in adenomyosis tissues (47.37%) was significantly lower than that in eutopic endometrium (71.05%) and normal endometrium (96.67%, P <0.0125). The expression of Survivin and E-cadherin in adenomyosis lesions showed a linear negative correlation (r = -0.988, P <0.01). Western blot results showed that the relative expression of Survivin protein in adenomyosis lesions was significantly higher than that in normal endometrium, E- The relative expression of cadherin in adenomyosis lesions was significantly lower than that in normal endometrium. Conclusion Survivin and E-cadherin may be related to the occurrence and development of adenomyosis.
