目的:研究三叶青水提物对慢性阻塞性肺疾病(COPD)大鼠白细胞介素23(IL-23)和白细胞介素17(IL-17)的影响及其作用机制。方法:清洁级健康雄性SD大鼠共30只,将其随机分成对照组、模型组和三叶青组,每组10只。利用烟熏加气管内滴入脂多糖(LPS)复制COPD模型,第8天开始用三叶青水提物进行干预,28天后采集标本,观察各组大鼠肺组织的病理变化,酶联免疫吸附试验(ELISA)检测血清、肺组织及支气管肺泡灌洗液(BALF)中IL-23和IL-17的表达情况。结果:血清中IL-23和IL-17的表达分别为模型组(25.38±1.25)ng/L、(63.78±4.98)ng/L,对照组(10.23±0.35)ng/L、(40.28±3.08)ng/L,三叶青组(15.79±2.38)ng/L、(49.97±3.65)ng/L,三组两两比较差异均有统计学意义(P<0.05)。肺组织匀浆中的IL-23和IL-17表达分别为模型组(45.83±2.22)ng/L、(90.68±0.12)ng/L,对照组(16.39±1.14)ng/L、(56.23±1.24)ng/L,三叶青组(30.89±6.13)ng/L、(70.79±0.08)ng/L,三组两两比较差异均有统计学意义(P<0.05)。BALF中的IL-23和IL-17表达分别为模型组(35.83±3.22)ng/L、(83.34±4.35)ng/L,对照组(14.68±1.14)ng/L、(52.17±3.92)ng/L,三叶青组(24.89±1.13)ng/L、(62.65±2.23)ng/L,三组两两比较差异均有统计学意义(P<0.05)。结论:三叶青水提物可降低COPD大鼠血清、肺组织及支气管肺泡灌洗液中IL-23和IL-17的表达,对COPD炎症的防治有重要的意义。
Objective: To investigate the effects and mechanisms of aqueous extract of clover leaf on interleukin-23 (IL-23) and interleukin-17 (IL-17) in chronic obstructive pulmonary disease (COPD) rats. Methods: Thirty healthy male SD rats were randomly divided into control group, model group and clover leaf group, with 10 rats in each group. The model of COPD was established by using lipopolysaccharide (LPS) dripped in the flue gas-filled tube. The aqueous extract of Clover was used to intervene on the 8th day. After 28 days, the specimens were collected to observe the pathological changes of the lung tissue in each group. Enzyme-linked immunosorbent assay The levels of IL-23 and IL-17 in serum, lung tissue and bronchoalveolar lavage fluid (BALF) were detected by ELISA. Results: The levels of IL-23 and IL-17 in serum were (25.38 ± 1.25) ng / L, (63.78 ± 4.98) ng / L and 10.23 ± 0.35 ) ng / L, and Clover group (15.79 ± 2.38) ng / L, (49.97 ± 3.65) ng / L respectively. There was significant difference between the two groups (P <0.05). The expression of IL-23 and IL-17 in lung homogenate were (45.83 ± 2.22) ng / L and (90.68 ± 0.12) ng / L in the model group, 16.39 ± 1.14 and 56.23 ± 1.24) ng / L, Clover group (30.89 ± 6.13) ng / L, (70.79 ± 0.08) ng / L respectively. There was significant difference between the two groups (P <0.05). The expression of IL-23 and IL-17 in BALF were (35.83 ± 3.22) ng / L and (83.34 ± 4.35) ng / L in model group and (14.68 ± 1.14) ng / L and / L, clover leaf group (24.89 ± 1.13) ng / L, (62.65 ± 2.23) ng / L, the difference between the two groups was statistically significant (P <0.05). Conclusion: Clover water extract can reduce the expression of IL-23 and IL-17 in serum, lung tissue and bronchoalveolar lavage fluid of COPD rats, which is of great significance for the prevention and treatment of COPD inflammation.
