目的建立HPLC-ELSD法测定丹参及其提取物中Na、K元素的方法。方法色谱柱为Waters Sepherisorb SCX柱(250 mm×4.6 mm,5μm);流动相为0.05 mol/L醋酸铵(醋酸调节pH值至4.8)-乙腈(70∶30),体积流量为1.0 mL/min,柱温30℃;ELSD漂移管温度100℃,载气体积流量2 L/min,检测器增益为2。结果 Na元素在19.7~197.0μg/mL(r=0.999 8)线性关系良好,K元素在35.0~350.0μg/mL(r=0.999 5)线性关系良好;Na、K平均回收率分别为99.0%、100.2%,RSD分别为0.7%、0.8%。结论所建立的方法简单可靠,可以用于测定丹参及其提取物中Na、K元素。
Objective To establish a HPLC-ELSD method for the determination of Na and K in Salvia miltiorrhiza and its extracts. Methods The column was a Waters Sepherisorb SCX column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of 0.05 mol / L ammonium acetate (acetic acid adjusted to pH 4.8) - acetonitrile (70:30) and the volume flow rate was 1.0 mL / min , Column temperature 30 ℃, ELSD drift tube temperature 100 ℃, carrier gas volume flow 2 L / min, detector gain 2. Results The linearity of Na was good at 19.7 ~ 197.0μg / mL (r = 0.999 8). The linearity of K was 35.0 ~ 350.0μg / mL (r = 0.999 5). The average recoveries of Na and K were 99.0% 100.2% with RSD of 0.7% and 0.8% respectively. Conclusion The established method is simple and reliable and can be used to determine Na and K elements in Radix Salviae Miltiorrhizae and its extracts.
