采用戊二醛二步法将心钠素与牛血清白蛋白(BSA)偶联为大分子全抗原,免疫日本大耳白家兔,制备抗血清。用氯胺T法标记心钠素。被测血浆标本先在85℃中加热10min,离心后取上清液测定心钠素含量。本法批内CV6％,批间CV12.7％。应用RIA测定30例正常成年人血浆心钠素含量在110.48±45.25ng/L。风心病伴心力衰竭11例患者,其中8例心钠素值在160ng/L以上。
Two-step glutaraldehyde method was used to conjugate atrial natriuretic peptide and bovine serum albumin (BSA) as macromolecular antigens to immunize Japanese white rabbits to prepare antiserum. Atrial natriuretic factor was labeled with chloramine T method. Plasma samples were first measured at 85 ℃ heating 10min, centrifuged supernatant was measured atrial natriuretic peptide. CV6% within the law approved, between CV12.7%. The plasma atrial natriuretic peptide levels of 30 normal adults were measured by RIA at 110.48 ± 45.25ng / L. Rheumatic heart disease with heart failure in 11 patients, including 8 cases of atrial natriuretic peptide value of 160ng / L or more.
