目的:研究白杨素(ChR)是否具有增敏肿瘤坏死因子相关凋亡诱导配体(TRAIL)诱导人胃癌SGC-7901细胞凋亡作用。方法:体外培养SGC-7901细胞。碘化丙啶(PI)染色流式细胞术(FCM)分析细胞凋亡率,琼脂糖凝胶电泳检测DNA梯形条带。结果:ChR(40μmol/L)、TRAIL(100 ng/mL)以及两者合用的细胞凋亡率分别为(4.57±0.54)%、(3.70±0.17)%和(48.81±4.11)%。两者合用后,琼脂糖凝胶电泳呈现细胞凋亡的典型DNA梯形条带。结论:白杨素具有敏化TRAIL诱导人胃癌SGC-7901细胞凋亡作用。
AIM: To investigate whether chrysene (ChR) can induce the apoptosis of human gastric cancer cell line SGC-7901 by sensitizing tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). Methods: SGC-7901 cells were cultured in vitro. Propidium iodide (PI) staining flow cytometry (FCM) analysis of apoptotic rate, DNA ladder detection by agarose gel electrophoresis. Results: The rates of apoptotic cells were (4.57 ± 0.54)%, (3.70 ± 0.17)% and (48.81 ± 4.11)% respectively when treated with ChR (40μmol / L) and TRAIL (100ng / mL) After the combination of the two, a typical DNA ladder showing apoptotic cells was obtained by agarose gel electrophoresis. Conclusion: Chrysin can sensitize TRAIL to induce apoptosis of human gastric cancer SGC-7901 cells.
