目的探讨含survivin siRNA的PcDNA3-siRNA对裸鼠人乳腺癌MCF-7细胞移植瘤微血管生成的影响。方法制作裸鼠MCF-7细胞移植瘤模型,然后随机分为PcDNA3-siRNA处理组,PcDNA3处理组,空白对照组,成瘤后分别将PcDNA3-siRNA质粒,、PcDNA3空质粒,生理盐水注入裸鼠MCF-7移植瘤中。采用RT-PCR及Western blot半定量法检测各组移植瘤组织中survivin mRNA及survivin蛋白的表达;SP免疫组化法检测各组移植瘤中的微血管密度(MVD)。结果 PcDNA3-siRNA组survivin mRNA及survivin蛋白的表达量较PcDNA3组及空白对照组明显降低(P(0.05),且PcDNA3-siRNA组MVD明显低于PcDNA3组及空白对照组(P(0.05);PcDNA3组与空白对照组survivin mRNA,survivin蛋白及MVD值差异无统计学意义(P(0.05)。结论 PcDNA3-siRNA可抑制裸鼠MCF-7细胞移植瘤survivin的表达,而且对移植瘤的微血管生成具有明显的抑制作用。
Objective To investigate the effect of PcDNA3-siRNA containing survivin siRNA on the microvascular angiogenesis of human breast cancer MCF-7 cells in nude mice. Methods The nude mouse model of MCF-7 cell xenografts was established and then randomly divided into PcDNA3-siRNA group, PcDNA3-treated group and blank control group. PcDNA3-siRNA plasmid, PcDNA3 empty plasmid and normal saline were injected into nude mice MCF-7 xenografts. The expression of survivin mRNA and survivin protein were detected by semi-quantitative RT-PCR and Western blot in each group. The MVD in each group was detected by SP immunohistochemistry. Results The expression of survivin mRNA and survivin protein in PcDNA3-siRNA group was significantly lower than that in PcDNA3 group and blank control group (P <0.05), and the MVD in PcDNA3-siRNA group was significantly lower than that in PcDNA3 group and blank control group (P <0.05) There was no significant difference in survivin mRNA, survivin protein and MVD between group and blank control group (P (0.05) .Conclusion PcDNA3-siRNA can inhibit the expression of survivin in MCF-7 cell xenograft in nude mice, Obvious inhibition.
