目的:了解Th17细胞、Treg细胞特异性转录因子RORγt (RORγt mRNA)、Foxp3(Foxp3 mRNA)在急性期玫瑰糠疹(PR)患者外周血单个核细胞中的表达水平,探讨其在玫瑰糠疹急性期发病中的作用.方法:采用荧光实时定量反转录聚合酶链反应(RT-PCR)检测20例玫瑰糠疹急性期患者(PR组)和20例健康体检者(健康对照组)外周血单个核细胞中RORγt和Foxp3mRNA的表达水平.结果:PR组RORγt mRNA、Foxp3 mRNA的表达水平均显著高于健康对照组,差异有统计学意义(t值分别是3.140、2.516,P均<0.05);PR组RORγt/Foxp3 mRNA表达水平的比值(2.31±0.90)增大,与健康对照组(0.86±0.40)比较,差异有统计学意义(t=4.445,P<0.01).PR组RORγt mRNA与Foxp3 mRNA的表达水平呈正相关(r=0.811,P<0.01);健康对照组RORγt mRNA与Foxp3 mRNA表达水平无相关性(r=-0.253,P>0.05).RORγt mRNA表达水平与玫瑰糠疹严重程度评分(18.33±7.56)呈正相关(r=0.634,P<0.05),Foxp3 mRNA表达水平与玫瑰糠疹严重程度评分(18.33±7.56)无相关性(r=0.171,P>0.05).结论:玫瑰糠疹患者外周血RORγt mRNA与Foxp3 mRNA的表达水平均显著增高,且以RORγt mRNA的表达占优.Th17/Treg细胞比例失衡,可能在玫瑰糠疹的发病中起着一定的作用.“,”Objective:To investigate the expression levels of h17 and Treg cell specific transcription factors,RORγt and Foxp3 mRNA,in peripheral blood mononuclear cells(PBMCs) of patients with acute pityriasis rosea(PR) and their roles in the pathogenesis of acute PR.Methods:Peripheral venous blood were obtained from 20 patients with PR in acute stage(PR group) and 20 healthy humans(control group).Real-time fluorescence-based quantitative polymerase chain reaction(RT-PCR) was carried out to measure the expressions levels of RORγt and Foxp3 mRNA in PBMCs.Results:The relative expression levels of RORγt and Foxp3 mRNA were significantly higher in PR than in the controls(t=3.140,P<0.05;t=2.516,P<0.05).The ratios of RORγt to Foxp3 mRNA in two groups were 2.31-±0.90 and 0.86-±0.40,respectively,(t=4.445,P<0.01).The expression levels of RORγt mRNA was positively correlated with that of Foxp3 mRNA in PR group(r=0.811,P<0.05),but not in the controls(r=-0.253,P>0.05).The severity scores of pityriasis rosea(18.33±7.56) was positively correlated with the expression levels of RORγt mRNA(r=0.634,P<0.05),but not Foxp3 mRNA(r=0.171,P9>0.05).Conclusions:The expression levels of RORγt and Foxp3 mRNA are significantly higher in acute stage of PR,and expression of RORγt mRNA is dominant.The imbalance of Th17/Treg cells in PBMCs could plays pathogenic role in acute pityriasis rosea.
