研究热水浸提杏鲍菇中粗多糖的提取工艺路线,多糖含量用苯酚-硫酸法测得。在单因素试验的基础上,选定料液比、提取时间及醇沉浓度3个因素的3个水平进行中心组合试验,建立了多糖含量的二次回归方程,进一步优化了提取工艺条件,验证了模型的有效性,通过响应面分析以及岭嵴分析得到了优化组合条件。试验结果表明,醇沉浓度、提取时间对多糖得率都有显著影响,而料液比没有显著影响。当提取工艺条件为提取时间2.5h,料液比1∶38和醇沉浓度为91%时,水提杏鲍菇多糖得率达到极大值。此条件下多糖得率预测值为6.58%,验证值为6.52%。
The extraction process of crude polysaccharides in hot water extract of Pleurotus eryngii was studied. Polysaccharide content was measured by phenol-sulfuric acid method. On the basis of single factor experiment, three combinations of three factors were selected, such as the ratio of solid to liquid, extraction time and alcohol concentration. The quadratic regression equation of polysaccharide content was established and the extraction process conditions were further optimized. The effectiveness of the model was verified by response surface analysis and Ridge ridge analysis. The results showed that alcohol concentration and extraction time had a significant effect on the yield of polysaccharides, while the feed-liquid ratio had no significant effect. When the extraction process conditions for the extraction time 2.5h, solid-liquid ratio of 1:38 and alcohol sediment concentration of 91%, the water extract Pleurotus eryngii polysaccharide yield reached its maximum. Under this condition, the predicted yield of polysaccharide was 6.58% and the verification value was 6.52%.
