目的比较燃煤型砷中毒患者与正常人群金属硫蛋白(MT)表达,研究金属硫蛋白基因在燃煤型砷中毒患者中的表达及其意义,阐明燃煤型砷中毒分子机制。方法燃煤型砷中毒患者与正常人群各50例,采用Trizol-酚-氯仿一步法提取总RNA。紫外分光光度法测定RNA的量,A260/A280鉴定RNA纯度。采用实时荧光RT-PCR技术测定燃煤型砷中毒MT mRNA的表达,以-βactin作为实时荧光RT-PCR的质控,当P<0.05时认为差异有统计学意义。结果正常组MT mRNA的相对表达水平为6.82±7.80,砷中毒组MT mRNA的相对表达水平为2.02±2.82。燃煤型砷中毒患者MTmRNA表达降低,约为正常对照的30%,二者间差异有统计学意义(P<0.01)。结论金属硫蛋白在燃煤型砷中毒代谢中有重要意义,起重要的调控作用。MT mRNA表达降低可能是砷中毒机制之一。
Objective To compare the expression of metallothionein (MT) in patients with arsenism with coal-fired arsenism and normal subjects, and to study the expression and significance of metallothionein in patients with arsenism of coal-burning arsenism. Methods Fifty patients with normal arsenic poisoning and coal were respectively treated with Trizol-phenol-chloroform method for one-step extraction of total RNA. The amount of RNA was determined by UV spectrophotometry and A260 / A280 was used to identify RNA purity. Real-time fluorescent RT-PCR was used to detect the expression of MT mRNA in coal-fired arsenic poisoning. The quality control of real-time fluorescent RT-PCR was -βactin. When P <0.05, the difference was statistically significant. Results The relative expression level of MT mRNA in normal group was 6.82 ± 7.80 and that of arsenism group was 2.02 ± 2.82. The MT mRNA expression in patients with coal-burning arsenism was decreased, which was about 30% of the normal control, the difference was statistically significant (P <0.01). Conclusion Metallothionein plays an important role in the metabolism of coal-borne arsenic poisoning, and plays an important regulatory role. Decreased MT mRNA expression may be one of the mechanisms of arsenic poisoning.
