分析了大麦黄花叶病毒的外壳蛋白及基因组RNA的性状。SDS-聚丙烯酰胺凝胶电泳表明,该病毒的外壳蛋白由单种亚基组成,其分子量为34.9KDa,在常规提纯过程中,该病毒的外壳蛋白较不稳定,部分易降解为28.0KDa和26.6KDa的组分;不同毒源病毒的外壳蛋白分子量无明显差异,但其稳定性差异较大。1.2%变性琼脂糖凝胶电泳的测定结果表明,大麦黄花叶病毒的基因组RNA由二组分组成,其大小分别为7.78Kb(RNA-1)和3.48Kb(RNA-2),且不同毒源间无明显差异。以Oligo(dT)15为引物反转录合成cDNA的实验表明,RNA-1和RNA-2的3端均带poly(A)的结构,与国外已报道的相符。
The barley yellow mosaic virus coat protein and genomic RNA traits were analyzed. SDS-polyacrylamide gel electrophoresis showed that the coat protein of the virus consists of a single subunit with a molecular weight of 34.9 kDa. During normal purification, the coat protein of the virus is less stable and partially degraded to 28. 0KDa and 26.6KDa components; different virulent virus shell protein molecular weight no significant difference, but its stability is quite different. The results of 1.2% denaturing agarose gel electrophoresis showed that the genomic RNA of barley yellow mosaic virus was composed of two components, which were 7.78Kb (RNA-1) and 3.48Kb (RNA-2) in size, There was no significant difference between different virulent sources. The experiment of reverse transcribed cDNA with Oligo (dT) 15 as primer showed that the 3 ’end of both RNA-1 and RNA-2 had a poly (A) structure, which was in accordance with the reported abroad.
