目的 通过检测多囊卵巢综合征 (PCOS)病人的胰岛素敏感指数等指标 ,观察是否存在胰岛素抵抗 ,对 PCOS病人胰岛素受体底物 - 1(IRS- 1)基因多态性进行分析 ,探讨 IRS- 1基因与 PCOS的关系 ,判断其可否作为PCOS胰岛素抵抗的致病基因。 方法 采用 PCR技术检测正常人与 PCOS病人 IRS- 1的等位基因与基因频率 ,据不同的基因型 (GG、GA或 AA)将病人分为 3组 ,比较各组的体重指数、腰臀围比例、黄体生成素、游离睾酮、雄烯二酮、胰岛素敏感指数的水平。 结果 多囊卵巢综合征不同的基因型各生化指标差异无显著性 ,但确实存在胰岛素抵抗 ,胰岛素敏感指数可较好地反映其程度。 结论 PCOS的发病与胰岛素受体底物 - 1基因变异无明显关系。
Objective To detect the insulin resistance in patients with polycystic ovary syndrome (PCOS) and to detect the polymorphism of insulin receptor substrate-1 (IRS-1) gene in PCOS patients and to explore whether IRS- 1 gene and PCOS to determine whether it can be used as a causal agent of PCOS insulin resistance. Methods The allele and gene frequency of IRS-1 in normal and PCOS patients were detected by PCR. The patients were divided into 3 groups according to different genotypes (GG, GA or AA). Body mass index, waist-hip circumference Ratio, luteinizing hormone, free testosterone, androstenedione, insulin sensitivity index. Results There was no significant difference in the biochemical indexes between different genotypes of polycystic ovary syndrome. However, insulin resistance did exist, and insulin sensitivity index could reflect its degree. Conclusion The incidence of PCOS has no significant relationship with the variation of insulin receptor substrate - 1 gene.
