目的:对右旋糖酐40甘露醇注射剂的药物动力学和药效进行分析。方法:选取实验中心饲养的16只家兔为A组,将A组家兔分为两个亚组(1组、2组),每组8只,先后分别注射右旋糖酐40甘露醇注射剂和甘露醇注射液。对复合剂的动力学特点进行分析。同时另选18只家兔为B组,同样分为两个亚组(1组、2组),禁食后分别注射甘露醇和右旋糖酐40,而后抽取家兔不同时间段血液,对两种药物的药物动力学和药效进行调查。结果:A组家兔体内甘露醇和右旋糖酐甘露醇的药物浓度相似,两组药物动力学参数比较无差异。B组家兔在注射药物后0.25h、8h比较中排尿量无明显差异;但在注射药物有0.5h、1h、2h、4h比较中,排尿量存在明显差异,说明甘露醇的利尿效果要好于右旋糖酐。结论:右旋糖酐40联合甘露醇使用效果要好于单方用药的效果,联合用药后药物起效时间明显缩短,能够达到快速治疗的目的。
Objective: To analyze the pharmacokinetics and pharmacodynamics of dextran 40 mannitol injection. Methods: Totally 16 rabbits raised in the experimental center were divided into group A and divided into two subgroups (group 1 and group 2), each group consisting of 8 rabbits. The dextran 40 mannitol and mannitol Injection. The dynamic characteristics of the complex were analyzed. At the same time, another 18 rabbits were selected as Group B, and divided into two subgroups (Group 1 and Group 2). After fasting, mannitol and dextran 40 were respectively injected, and then blood was collected from rabbits at different time intervals. Pharmacokinetics and efficacy were investigated. Results: The drug concentration of mannitol and dextran mannitol in rabbits in group A were similar, and there was no difference in the pharmacokinetic parameters between the two groups. In group B, there was no significant difference in urinary output between 0.25h and 8h after injection, but there was a significant difference in urine output between 0.5h, 1h, 2h and 4h after injecting drugs, indicating that diuretic effect of mannitol is better than Dextran. Conclusion: Dextran 40 combined with mannitol is better than unilateral medication, and the onset time of the drug is obviously shortened after combination therapy, which can achieve the purpose of rapid treatment.
