目的 :研究腺病毒 (adenovirus ,Ad) 2型E1B 5 5KD癌蛋白 (Ad2E1B 5 5KD)与人Daxx(humanDaxx ,hDaxx)的相互作用及作用部位。方法 :利用共免疫沉淀反应研究Ad2 / 5E1B 5 5KD与hDaxx的结合反应 ,通过酵母双杂交体系测定两种蛋白质的相互作用。结果 :酵母双杂交试验结果显示Ad2E1B 5 5KD通过C端 5 8个氨基酸 (aminoacid ,aa)与hDaxx结合并发生相互作用。共免疫沉淀反应和Westernblot结果证实Ad2 / 5E1B 5 5KD能在细胞内外与hDaxx直接结合。结论 :Ad2E1B 5 5KD与hDaxx结合并发生相互作用。
Objective: To investigate the interaction and site of adenovirus (Ad) type 2 E1B5 5KD oncoprotein (Ad2E1B 5 5KD) with human Daxx (human Daxx, hDaxx). Methods: The co-immunoprecipitation assay was used to study the binding of Ad2 / 5E1B 5 5KD to hDaxx and the interaction between the two proteins was determined by yeast two-hybrid system. Results: The result of yeast two-hybrid assay showed that Ad2E1B 5 5KD interacted with hDaxx via C-terminal 58 amino acids (aminoacid, aa). Co-immunoprecipitation and Western blot confirmed that Ad2 / 5E1B 5 5KD could directly bind to hDaxx in vitro and in vivo. Conclusion: Ad2E1B 5 5KD binds to hDaxx and interacts with it.
