目的筛选肾综合征出血热滴鼻疫苗壳聚糖载体,为进一步研究其滴鼻或气雾疫苗提供依据。方法 48只昆明鼠按完全随机分组的方法分为4组,每组12只,采用3种不同粘度的壳聚糖作为载体,制成终浓度为0.1%壳聚糖的病毒液进行鼻腔接种,采用ELISA方法检测肾综合征出血热特异性Ig G抗体。结果疫苗接种后,小鼠能够对各剂量组产生免疫反应,Ⅱ、Ⅲ组能诱导较高、较快血清抗体滴度的增加,免疫后3周,血清抗体滴度维持在较高的水平,但各个实验组间无统计学差异(P>0.05)。结论本实验得出壳聚糖能够增强疫苗的抗体反应,Ⅱ、Ⅲ组效果更佳。
Objective To screen chitosan carrier of hemorrhagic fever with intranasal vaccine for intranasal hemorrhagic fever with nasal syndrome in order to provide a basis for further study on its intranasal or aerosol vaccine. Methods 48 Kunming mice were randomly divided into 4 groups with 12 mice in each group. Three kinds of chitosan with different viscosity were used as the carrier to make the nasal inoculation of the virus liquid with the final concentration of 0.1% chitosan. ELISA method for detection of hemorrhagic fever with renal syndrome specific Ig G antibody. Results After vaccination, the mice were able to produce an immune response to each dose group. In groups Ⅱ and Ⅲ, the antibody titers of higher and faster serum were increased. After three weeks of immunization, the serum antibody titer was maintained at a high level, However, there was no significant difference among the experimental groups (P> 0.05). Conclusion This experiment shows that chitosan can enhance the antibody response of the vaccine, and the effect of group Ⅱ and Ⅲ is better.
