目的:研究分析CXCLl及其受体CXCR2在胃癌组织中的表达情况,探讨CXCLl及其受体CXCR2在胃癌发生和浸润转移中的作用及临床意义。方法:采用免疫组织化学染色及Western blot检测40例胃癌患者正常组织及癌组织中CXCL1及CX-CR2蛋白表达,对各表达情况与不同临床病理特征进行统计学分析,采用逆转录-聚合酶链反应(reverse transcription-polymnerase chain reaction,RT-PCR)方法检测19例CXCL1在mRNA水平的表达情况。结果:在胃癌及转移淋巴结组织中CXCLl及CXCR2在蛋白及mRNA水平均高表达,且与临床分期相关(P<0.05)。结论:CXCL1及CXCR2在胃癌组织中的高表达与胃癌的发生、发展及侵袭转移相关,可能在胃癌淋巴转移过程中起重要作用。
OBJECTIVE: To study the expression of CXCL1 and its receptor CXCR2 in gastric cancer and to explore the role and clinical significance of CXCL1 and its receptor CXCR2 in gastric carcinogenesis and invasion and metastasis. Methods: Immunohistochemical staining and Western blot were used to detect the expression of CXCL1 and CXCR2 in normal tissues and cancerous tissues of 40 patients with gastric cancer. The expressions of CXCL1 and CXCR2 were detected by immunohistochemical staining and Western blot respectively. (RT-PCR) was used to detect the expression of CXCL1 in 19 cases at mRNA level. Results: The expressions of CXCL1 and CXCR2 in gastric cancer and metastatic lymph nodes were both high in both protein and mRNA levels, and were correlated with clinical stage (P <0.05). Conclusion: The high expression of CXCL1 and CXCR2 in gastric cancer is associated with the occurrence, development, invasion and metastasis of gastric cancer and may play an important role in the lymphatic metastasis of gastric cancer.
