目的:探讨糖肾方对自发性2型糖尿病模型OLETF大鼠肾脏的保护作用以及对血中白细胞介素-10、肿瘤坏死因子-α和多巴胺的影响。方法:OLETF大鼠随机分为模型组、糖肾方组(1.6g/kg)、蒙诺组(1.0mg/kg)、LETO大鼠为正常对照组。各组动物从12周龄起进行灌胃给药,连续给药44周。实验结束,大鼠56周龄时,测定血糖和24h尿蛋白水平、血清中白细胞介素-10、肿瘤坏死因子-α、多巴胺。取肾组织观察肾组织形态学改变,并对肾损害进行半定量评分。结果:糖肾方可降低模型组大鼠血中TNF-α、DA水平,升高IL-10水平。降低肾小球硬化指数和肾小管间质纤维化指数,改善肾组织形态学异常。结论:糖肾方对2型糖尿病模型OLETF大鼠肾脏具有保护作用,延缓其病理进展,其作用机制可能与调节体内炎症因子水平有关。
Objective: To investigate the protective effects of Tangshen Formula on kidney of OLETF rats and its effect on interleukin-10, tumor necrosis factor-α and dopamine in blood of spontaneously type 2 diabetic rats. Methods: OLETF rats were randomly divided into model group, Tangshen Formula group (1.6g / kg), Monopril group (1.0mg / kg) and LETO rats as normal control group. The animals in each group were given gavage from the age of 12 weeks for continuous administration for 44 weeks. At the end of the experiment, blood glucose, 24h urinary protein level, serum interleukin-10, tumor necrosis factor-alpha and dopamine were measured at 56 weeks of age. Kidney tissue was taken to observe the morphological changes of the kidney, and renal damage were semi-quantitative score. Results: Tangshen Formula can reduce the levels of TNF-α and DA in the blood of model rats and increase the level of IL-10. Reduce glomerular sclerosis index and tubulointerstitial fibrosis index, improve renal morphological abnormalities. Conclusion: Tangshen formula has a protective effect on the kidney of OLETF rat model of type 2 diabetes and slows its pathological progress. Its mechanism may be related to the regulation of inflammatory cytokines in vivo.
