采用混料设计,研究混合酶配比,优化水解条件;HPCE法测定血管紧张素转换酶(ACE)抑制率。研究混合酶法制备紫菜蛋白降压肽的优化工艺。建立混合酶比例与降压活性的数学模型,确定最佳酶配比:Alcalase碱性蛋白酶38.1%,Neutrase中性蛋白酶31.9%,胰蛋白酶30.0%。最佳水解条件:底物质量分数5%、加酶量3%、pH 8.5、温度50℃、时间90 min。混合酶较单一酶制备降压肽的ACE抑制率提高了44.33%~86.45%。
Mixture design was used to study the mixture ratio of mixed enzymes to optimize the hydrolysis conditions; HPCE method was used to measure the inhibition rate of angiotensin converting enzyme (ACE). Study on the optimization of preparation of seaweed protein antihypertensive peptide by mixed enzyme method. The mathematical model of mixed enzyme activity and antihypertensive activity was established, and the best enzyme ratio was determined: Alcalase alkaline protease 38.1%, Neutrase neutral protease 31.9% and trypsin 30.0%. The optimum hydrolysis conditions: substrate mass fraction 5%, enzyme 3%, pH 8.5, temperature 50 ℃, time 90 min. Compared with single enzyme preparation, the ACE inhibitory rate of the hypotensive peptides increased 44.33% ~ 86.45%.
