目的:建立乌梅药材中多糖的含量测定方法。方法:采用苯酚-硫酸显色法测定乌梅药材中多糖的含量。结果:样品经苯酚-硫酸显色后于488 nm处测定吸收度,在此波长处溶液的吸收度与葡萄糖含量呈良好的线性关系;线性范围0.02~0.2 g·L-1(r=0.999 8),加样回收率100.38%,RSD 0.892%;乌梅中多糖含量2.51%。结论:苯酚-硫酸显色法操作简便,结果准确,为乌梅多糖的进一步研究奠定良好的基础。
Objective: To establish a method for the determination of polysaccharide in Fructus Junmasensis. Methods: The content of polysaccharides in Fructus Mume was determined by phenol - sulfuric acid colorimetry. Results: The absorbance at 488 nm was determined by phenol-sulfuric acid spectrophotometry. The absorbance of the solution showed a good linear relationship with the glucose content. The linear range was 0.02-0.2 g · L-1 (r = 0.999 8) ), The recovery rate was 100.38%, the RSD was 0.892%. The polysaccharide content in plum was 2.51%. Conclusion: Phenol-sulfuric acid colorimetric method is simple and convenient, the results are accurate, and lay a good foundation for the further study of Polysaccharide.
