目的探讨端粒酶逆转录酶(hTERT)、c-myc、cyclin D1基因在胶质瘤中的表达与预后的关系。方法收集56例手术切除的胶质瘤标本,应用原位杂交技术和免疫组化S-P法分别检测胶质瘤组织中hTERT和c-myc、cyclin D1基因的表达;6例正常脑组织作为对照。并对检测结果与随访结果进行综合分析。结果56例胶质瘤中,30例表达hTERT,阳性率为53.6%;33例表达c-myc基因,阳性率为58.9%;32例表达cyclin D1基因,阳性率为57.1%。正常脑组织不表达hTERT、c-myc、cyclin D1。随着胶质瘤分级的升高,hTERT和c-myc、cyclin D1基因表达增强(P<0.05)。hTERT与c-myc、cyclin D1基因的表达两两成正相关关系(P<0.05),三者的表达均与患者生存时间成负相关关系(P<0.01)。结论hTERT、c-myc、cyclin D1基因的过表达与胶质瘤细胞的恶性变和过度增殖有关,三者联合检测可作为临床判断胶质瘤患者预后的重要指标。
Objective To investigate the relationship between the expression of telomerase reverse transcriptase (hTERT), c-myc and cyclin D1 in gliomas and its prognosis. Methods 56 cases of glioma were collected. The expression of hTERT, c-myc and cyclin D1 in glioma tissues were detected by in situ hybridization and immunohistochemical S-P method. Six normal brain tissues were used as control. And a comprehensive analysis of test results and follow-up results. Results Among 56 cases of gliomas, 30 cases expressed hTERT, the positive rate was 53.6%. 33 cases expressed c-myc gene, the positive rate was 58.9%. 32 cases expressed cyclin D1 gene, the positive rate was 57.1%. Normal brain tissue does not express hTERT, c-myc, cyclin D1. With the increase of glioma grade, the expression of cyclin D1 gene of hTERT and c-myc increased (P <0.05). There was a positive correlation between hTERT and c-myc, cyclin D1 gene expression (P <0.05). The expression of hTERT was negatively correlated with patient’s survival time (P <0.01). Conclusion The overexpression of hTERT, c-myc and cyclin D1 is associated with malignant transformation and hyperproliferation of glioma cells. Combined detection of hTERT, c-myc and cyclin D1 may be used as an important index to evaluate the prognosis of patients with glioma.
