近来研究已证实引起假体松动的重要原因是巨噬细胞与移植物相互作用所致的骨质吸收。巨噬细胞本身不能吸收骨质,然而它能分泌100种以上物质,其中有的能刺激骨吸收,有的能抑制骨吸收。比较重要的刺激因子有前列腺素(PG)、细胞素、转化生长因子、集落刺激因子和酶,而高剂量PGE_2有直接抑制作用,消炎痛亦会抑制骨质吸收。本文旨在阐述在这种相互作用中由巨噬细胞释放的递质,确定它们是刺激因子还是抑制因子,以及它们的性质。取下预先经~(45)Ca标记的4日龄TuckTO小鼠的颅骨,分成两片,每片包含部分额骨与顶骨,添加不同浓度的巨噬细胞后进行孵育,以释放到培养基中的~(45)Ca百分浓度作为骨吸收指
Recently, it has been confirmed that the major cause of loosening of the prosthesis is the bone resorption caused by the interaction between macrophages and the graft. Macrophages themselves can not absorb bone, however, it can secrete more than 100 substances, some of which can stimulate bone resorption, and some can inhibit bone resorption. The more important stimulating factors are prostaglandin (PG), cytokines, transforming growth factor, colony stimulating factor and enzymes, while high-dose PGE 2 has a direct inhibitory effect, and indomethacin also inhibits bone resorption. This article aims to elucidate the neurotransmitters released by macrophages in this interaction, determining whether they are stimulatory or inhibitory, and their properties. The skull of 4-day-old TuckTO mice previously labeled with ~ (45) Ca was removed and divided into two pieces, each piece containing part of the frontal and parietal bones.
