目的:探讨依曲替酸通过上调Fas基因表达诱导皮肤鳞状细胞癌(鳞癌)细胞系SCL-1细胞凋亡的作用。方法:以体外培养的人皮肤鳞癌细胞系SCL-1为研究对象,MTT法观察依曲替酸对SCL-1细胞的生长抑制作用,Annxin V-FITC/PI法检测早期凋亡,流式细胞仪检测细胞膜表面Fas的表达情况,实时荧光定量RT-PCR方法检测Fas mRNA的表达。结果:依曲替酸能显著抑制SCL-1细胞的生长,并具有浓度及时间依赖性。浓度为1×10-5mol/L的依曲替酸能诱导凋亡,上调细胞膜表面Fas表达水平及Fas mRNA水平。结论:依曲替酸能抑制皮肤鳞癌细胞生长及诱导凋亡,上调Fas抗原表达可能是其分子机制之一。
Objective: To investigate the role of ettringit acid in inducing apoptosis of SCL-1 cells in squamous cell carcinoma (SCC) by up-regulating Fas gene expression. Methods: The human skin squamous cell carcinoma cell line SCL-1 cultured in vitro was used as the research object. MTT assay was used to observe the inhibition of the growth of SCL-1 cells by MTT assay. The early apoptosis was detected by the flow cytometry Cytometry was used to detect the expression of Fas on the surface of cell membrane, and the expression of Fas mRNA was detected by real-time fluorescence quantitative RT-PCR. Results: It could significantly inhibit the growth of SCL-1 cells in a concentration-and time-dependent manner. The concentration of 1 × 10-5mol / L of triptorelin acid can induce apoptosis, increase the expression of Fas on the cell membrane surface and Fas mRNA levels. CONCLUSION: It can be one of the molecular mechanisms that it can inhibit the growth and induce the apoptosis of squamous cell carcinoma.
