目的:探讨亮叶杨桐总黄酮对高糖高脂膳食诱导的胰岛素抵抗大鼠脂肪细胞细胞信号转导抑制因子3(SOCS-3)和抵抗素过表达的抑制作用。方法:大鼠随机分为正常对照组、模型对照组和亮叶杨桐总黄酮400、200、100 mg·kg~(-1)·d~(-1)组,首先以高糖高脂膳食饲喂8周建立胰岛素抵抗大鼠模型,随后灌胃给药并继续高糖高脂膳食饲喂4周。于第12周末检测大鼠空腹血糖、胰岛素与抵抗素水平,计算稳态模型胰岛素抵抗指数(HOMA-IRI)的自然对数值,分别以实时荧光RT-PCR与流式细胞术检测脂肪细胞SOCS-3、抵抗素的表达。结果:与正常对照组比较,模型组脂肪细胞SOCS-3、抵抗素过表达,伴空腹血糖、胰岛素与抵抗素水平明显升高。与模型组比较,400,200 mg·kg~(-1)·d~(-1)亮叶杨桐总黄酮组的脂肪细胞SOCS-3、抵抗素表达水平显著下调,伴空腹血糖、胰岛素与抵抗素水平明显降低。结论:亮叶杨桐总黄酮可抑制胰岛素抵抗大鼠脂肪细胞SOCS-3和抵抗素过表达,从而降低空腹血糖、胰岛素与抵抗素水平。
OBJECTIVE: To investigate the inhibitory effects of total flavonoids of Litsea tung tree on fatty acid signal transduction inhibitory factor 3 (SOCS-3) and resistin over-expression in insulin resistance rats induced by high glucose and high fat diet. Methods: The rats were randomly divided into normal control group, model control group and Astragalus total flavonoids 400, 200, 100 mg · kg -1 · d -1 group. Firstly, high glucose and high fat diet Fed for 8 weeks to establish insulin resistance rat model, followed by intragastric administration and continued high-sugar high-fat diet for 4 weeks. Fasting plasma glucose, insulin and resistin levels were measured at the end of 12th week to calculate the natural logarithm of homeostasis model insulin resistance index (HOMA-IRI). Real-time fluorescent RT-PCR and flow cytometry were used to detect the SOCS- 3, resistin expression. Results: Compared with the normal control group, the expression of SOCS-3, resistin and fasting blood glucose, insulin and resistin in adipocytes of model group were significantly increased. Compared with the model group, the expression of resistin in adipocytes of 400,200 mg · kg -1 · d -1 flavonoids group was significantly down-regulated, accompanied with fasting blood glucose, insulin and resistin Significantly lower the level. CONCLUSION: The total flavonoids of Litsea velutina can inhibit the overexpression of SOCS-3 and resistin in adipocytes of insulin-resistant rats, so as to reduce the fasting blood glucose, insulin and resistin levels.
