目的 构建霍乱弧菌抗菌抗毒口服活疫苗候选株 ,并在家兔模型中评价它们的免疫原性及主动保护力。方法 应用染色体 质粒致死平衡系统 ,在无毒的霍乱弧菌中表达霍乱毒素基因 ,继而在家兔模型中进行免疫原性的比较。结果 构建的分别含有突变无毒的ctx 和ct B基因的IEM10 4和IEM10 5菌株免疫家兔后 ,刺激机体产生抗菌与抗毒抗体 ,前者的特异性抗体滴度较后者高 ,家兔主动保护试验中对纯霍乱毒素 (CT)攻击保护前者也高于后者。结论 构建的霍乱疫苗候选株IEM10 4和IEM10 5在家兔模型中均能产生较好的抗菌与抗毒免疫 ,IEM10 4对家兔的免疫原性和主动保护力略高于IEM10 5。
Objective To construct a candidate strain of Vibrio cholera antimicrobial and anti-virus live oral vaccine and evaluate their immunogenicity and active protection in rabbit model. Methods Chromosomal plasmid lethality balance system was used to express cholera toxin gene in non-toxic Vibrio cholerae, then the immunogenicity was compared in rabbit model. Results The constructed IEM10 4 and IEM10 5 strains with mutated and non-toxic ctx and ct B genes, respectively, stimulated the body to produce antibacterial and anti-toxin antibodies. The former had higher specific antibody titers than the latter, The protection test against pure cholera toxin (CT) attack protection is also higher than the former. Conclusion The cholera vaccine candidates IEM10 4 and IEM10 5 can produce good antibacterial and anti-toxin immunity in rabbit model. The immunogenicity and active protection of IEM10 4 to rabbits are slightly higher than that of IEM10 5.
