目的探讨caspase-12在偏侧咀嚼大鼠髁突软骨细胞中的表达变化。方法选取96只3周龄雄性SD大鼠,随机分为4个实验组和4个对照组。实验组大鼠拔除左侧上、下颌所有磨牙,对照组大鼠不做任何处理,分别于拔牙后1d,7d,14d,28d处死。免疫组化和RT-PCR方法检测不同时间点的caspase-12含量变化。结果拔牙后,实验组左侧的caspase-12表达在1d,7d,14d,28d时均显著高于对照组(P<0.05),实验组右侧的caspase-12表达在1d,7d,14d显著高于对照组(P<0.05)。随时间变化,实验组左侧caspase-12的含量逐渐升高,在14d,28d时升高较前一时间点有统计学差异(P<0.05),而实验组右侧caspase-12含量逐渐降低,在7d,28d时降低较前一时间点有统计学差异(P<0.05)。结论偏侧咀嚼导致双侧髁突软骨发生差异性改建,caspase-12参与了内质网应激介导的凋亡反应。
Objective To investigate the expression of caspase-12 in condylar chondrocytes of unilateral chewing rats. Methods 96 three-week-old male Sprague-Dawley rats were randomly divided into 4 experimental groups and 4 control groups. In the experimental group, all the molars on the left and lower jaws were removed, and the rats in the control group were sacrificed on the 1st, 7th, 14th and 28th day after tooth extraction respectively. Immunohistochemistry and RT-PCR were used to detect the changes of caspase-12 content at different time points. Results After tooth extraction, the expression of caspase-12 on the left side of the experimental group was significantly higher than that of the control group on the 1st, 7th, 14th and 28th days (P <0.05). The expression of caspase-12 on the right side of the experimental group was significantly higher on the 1st, Higher than the control group (P <0.05). The content of caspase-12 on the left side of the experimental group increased gradually with the passage of time, and the level of caspase-12 in the experimental group increased significantly at the 14th day and the 28th day (P <0.05), while the content of caspase-12 in the right side of the experimental group gradually decreased , And there was a significant difference at 7d and 28d when compared with the previous time point (P <0.05). Conclusions Unilateral chewing results in differential remodeling of bilateral condylar cartilage. Caspase-12 is involved in endoplasmic reticulum stress-mediated apoptosis.
