目的:探讨高剂量雌激素对新生SD大鼠前列腺形态发育的影响,及其与sonic hedgehog(SHH)信号通路之间的关系。方法:100只新生雄性仔鼠随机分为两组,出生后第0、1、3、5天分别给两组仔鼠皮下注射25μl玉米油和高剂量雌二醇(25μg/只,溶于25μl玉米油)。给药后于第1、6、10、20、30天取材前列腺,苏木精-伊红(HE)染色观察前列腺形态发育情况,免疫组织化学和RT-PCR分别检测SHH通路信号分子的表达。结果:高剂量雌激素组,新生SD大鼠前列腺腺体较小,腺管萎缩,发育明显受阻,在出生后第6、10、20、30天与对照组比较有显著差异;雌激素组SHHmRNA的表达在出生后第1、6、10、20、30天较对照组下调(P<0.05);雌激素组SHH信号通路的主要效应分子SHH、SMO、PTC和GLI-1的蛋白表达水平较对照组下调。结论:高剂量雌激素可能通过下调SHH信号通路,从而抑制新生SD大鼠前列腺的发育。
Objective: To investigate the effect of high-dose estrogen on prostatic hyperplasia in neonatal SD rats and its relationship with sonic hedgehog (SHH) signaling pathway. Methods: One hundred newborn male offspring were randomly divided into two groups. On the 0, 1, 3 and 5 days after birth, two groups of mice were injected subcutaneously with 25μl of corn oil and high dose of estradiol (25μg / Corn oil). Prostate was harvested on the 1st, 6th, 10th, 20th and 30th day after administration. Prostate morphology was observed by hematoxylin-eosin (HE) staining. The expression of SHH signaling pathway was detected by immunohistochemistry and RT-PCR. Results: In the high dose estrogen group, the newborn SD rats had smaller prostate glands and atrophy of the glandular duct. Obvious development was blocked. There was a significant difference between the high dose estrogen group and the control group on the 6th, 10th, 20th and 20th day after birth. The SHH mRNA (P <0.05). The expression of SHH, SMO, PTC and GLI-1 in the SHH signaling pathway of estrogen group was significantly lower than that of the control group on the 1st, 6th, 10th, 20th, Control group decreased. Conclusion: High-dose estrogen may inhibit the development of prostate in neonatal SD rats by down-regulating SHH signaling pathway.
