根据细菌磁颗粒对植物病毒颗粒的非特异性吸附作用,建立了一种新的提取植物病毒RNA的方法。利用细菌磁颗粒收集黄瓜绿斑驳花叶病毒、黄瓜花叶病毒和百合无症病毒等3种病毒颗粒,富集分离后,通过高温使其裂解释放出病毒RNA,进行RT-PCR检测。与利用Trizol试剂提取这3种植物病毒的RNA相比,具有操作简单、无毒性,能够显著缩短提取时间。对黄瓜绿斑驳花叶病毒的检测灵敏度验证表明:BMPs-RT-PCR与Trizol-RT-PCR的检测灵敏度相同,均为105倍的植物叶片组织稀释液。
Based on the nonspecific adsorption of bacterial virus particles by bacterial magnetic particles, a new method for extracting plant viral RNA was established. Bacterial magnetic particles were used to collect cucumber green mottle mosaic virus, cucumber mosaic virus and lily ectothermic virus and other three kinds of virus particles, after enrichment and separation, the virus RNA was lysed by pyrolysis and then detected by RT-PCR. Compared with RNA extracted with these three plant viruses by Trizol reagent, it is simple, non-toxic, and can shorten the extraction time significantly. The detection sensitivity of Cucumber green mottle mosaic virus showed that the detection sensitivity of BMPs-RT-PCR and Trizol-RT-PCR were the same, both of which were 105 times that of plant leaf tissue.
