目的:观察复方清热颗粒剂含药血清对超广谱β-内酰胺酶(ESBLs)的抑制作用。方法:采用紫外分光光度法,以空白血清、磷酸缓冲液为对照,以底物氨苄西林紫外吸光度(OD)变化为指标,观察不同浓度的中药复方清热颗粒剂含药血清对肺炎克雷伯杆菌ATCC700603产生的β内酰胺酶抑制作用。结果:复方清热颗粒剂33.33%含药血清对ATCC700603产β-内酰胺酶抑酶率超过50%,抑酶效果与血清浓度呈正相关(P<0.05),与对照组有显著性差异(P<0.01);结论:中药复方清热颗粒剂含药血清能够有效抑制ESBLs活性是其抗产ESBLs肺炎克雷伯杆菌急性感染的机制之一。
Objective: To observe the inhibitory effect of compound Qingre Granule drug-containing serum on extended-spectrum β-lactamase (ESBLs). METHODS: Ultraviolet spectrophotometry was used. Blank serum and phosphate buffer were used as controls. Substrate ampicillin ultraviolet absorbance (OD) changes were used as indicators to observe the effects of different concentrations of traditional Chinese medicine compound Qingre granules containing drug serum on Klebsiella pneumoniae. Beta lactamase inhibition produced by ATCC700603. RESULTS: The 33.33% drug-containing serum of compound Qingre granules produced more than 50% inhibition rate of β-lactamase produced by ATCC 700603. The effect of inhibiting enzyme and serum concentration was positively correlated (P<0.05), and there was significant difference from the control group (P< 0.01); Conclusion: The traditional Chinese medicine compound Qingre Granule containing serum can effectively inhibit the activity of ESBLs is one of the mechanisms of its anti-ESBLs Klebsiella pneumoniae acute infection.
