目的:探讨纤维蛋白原β链(Fgβ)-148C/T和-455G/A基因多态性与川崎病发病,及并发冠状动脉损伤(CAL)之间的关系。方法:应用聚合酶链反应-限制性内切酶片断长度多态性分析方法结合琼脂糖凝胶电泳技术,对47例川崎病患儿(川崎病组)和60例对照儿童(对照组)进行Fgβ-148C/T及-455G/A基因多态性分析。结果:①2组Fgβ-148C/T基因多态性的CC、CT、TT基因型频率和C、T等位基因频率均差异无统计学意义。川崎病组中,并发CAL者CT基因型频率明显高于未并发者(χ2=7.009,P<0.05)。②2组Fgβ-455G/A基因多态性的GG、GA、AA基因型频率和G、A等位基因频率均差异无统计学意义。川崎病组中,并发CAL者GA基因型频率明显高于未并发者(χ2=7.009,P<0.05)。结论:Fgβ-148C/T和Fgβ-455G/A基因的基因型和等位基因与川崎病的发生均无明显相关性。Fgβ-148C/T基因的CT基因型及Fgβ-455G/A基因的GA基因型与川崎病并发CAL的发生存在相关性。
Objective: To investigate the relationship between Fgβ -148C / T and -455G / A polymorphisms and the incidence of Kawasaki disease and coronary artery injury (CAL). Methods: 47 patients with Kawasaki disease (Kawasaki disease group) and 60 control children (control group) were studied by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and agarose gel electrophoresis Analysis of Fgβ-148C / T and -455G / A Gene Polymorphisms. Results: ① The frequency of genotypes CC, CT, TT and the frequency of C and T alleles in Fgβ-148C / T polymorphism in group 2 were not significantly different. In Kawasaki disease group, the frequency of CT genotype in patients with CAL was significantly higher than that in non-concurrent patients (χ2 = 7.009, P <0.05). ② The frequencies of GG, GA, AA genotypes and G, A allele frequencies of Fgβ-455G / A polymorphism in 2 groups were not significantly different. In Kawasaki disease group, the frequency of GA genotypes in patients with CAL was significantly higher than that in non-concurrent patients (χ2 = 7.009, P <0.05). Conclusion: The genotypes and alleles of Fgβ-148C / T and Fgβ-455G / A have no significant correlation with Kawasaki disease. The CT genotype of Fgβ-148C / T gene and the GA genotype of Fgβ-455G / A gene were correlated with the occurrence of Kawasaki disease complicated with CAL.
