2-氯乙胺盐酸盐化学改性壳聚糖制备的胺乙基壳聚糖(AECS),通过转谷氨酰胺酶催化与胶原蛋白肽(COP)接枝得到胺乙基壳聚糖-胶原蛋白肽(AECS-COP).采用单因素实验法得到AECS的最优工艺条件为:反应温度为80℃,反应时间为24h,2-氯乙胺盐酸盐与壳聚糖的质量比为2;得到AECS-COP的最优工艺条件为:反应温度40℃,反应时间1.2h,COP与AECS的质量比为1.2,转谷氨酰胺酶与AECS的质量比为0.13,该条件下的最大取代度为0.648.抗氧化实验和还原能力实验显示,AECS-COP具有很好的自由基清除能力和还原能力,且自由基清除能力和还原能力随其浓度和取代度的增加而增强.细胞实验数据表明,在0~1.0mg/L浓度范围内,AECSCOP的成纤维细胞增殖率随着浓度增加先增大后减小;在取代度为0~0.648范围内,AECS-COP的成纤维细胞增殖率随着取代度的增加而增大.
