该研究观察了毛蕊异黄酮对大鼠原代肝星状细胞(HSC)增殖和活化的影响,并探讨其是否通过影响核因子过氧化物酶体增殖因子活化受体γ(PPARγ)和法尼酯衍生物X受体(FXR)发挥作用。结果发现毛蕊异黄酮能抑制HSC的增殖,抑制活化标志物α-平滑肌肌动蛋白和Ⅰ型胶原的表达;随着HSC活化时间的延长,FXR表达逐渐降低,但毛蕊异黄酮对FXR表达没有明显影响;毛蕊异黄酮可浓度依赖性地上调PPARγ的表达及核转位,PPARγ拮抗剂可阻断毛蕊异黄酮对HSC活化的抑制作用。因此,结论认为毛蕊异黄酮可抑制HSC增殖和活化,其作用不通过FXR而是与上调PPARγ信号通路有关。
This study examined the effects of calycosin on the proliferation and activation of rat primary hepatic stellate cells (HSCs) and explored whether they affect PPARγ and farnesol by inhibiting the expression of nuclear factor peroxisome proliferator- Derivative X receptor (FXR) plays a role. The results showed that calycosin could inhibit the proliferation of HSC and inhibit the expression of α-smooth muscle actin and collagen Ⅰ. With the extension of HSC activation time, the expression of FXR decreased gradually, but the calycosin had no obvious effect on the expression of FXR. Calendula species could up-regulate the expression of PPARγ and nuclear translocation in a concentration-dependent manner. PPARγ antagonist could inhibit the inhibition of HSC activation by calycosin. Therefore, it was concluded that calycosin could inhibit the proliferation and activation of HSC, which is not related to FXR but upregulated PPARγ signaling.
