目的:探讨microRNA200c(miR-200c)和microRNA141(miR-141)在膀胱移行细胞癌中的表达及意义。方法:Mi-croRNA芯片筛选膀胱癌和癌旁组织差异表达的microRNA,实时定量RT-PCR法验证miR-200c和miR-141在膀胱癌组织和膀胱癌细胞系中的表达。结果:MiR-200c和miR-141的表达强度在膀胱肿瘤组织高于癌旁正常膀胱黏膜,在表浅性膀胱肿瘤高于肌层浸润性膀胱肿瘤,在高级别组膀胱肿瘤高于低级别组,在分化较好、恶性度较低的5637细胞系中表达较高,在分化较差、恶性度较高的T24、J82、EJ细胞系中表达均较低。结论:MiR-200c和miR-141可能参与膀胱癌的进展。
Objective: To investigate the expression and significance of microRNA200c (miR-200c) and microRNA141 (miR-141) in bladder transitional cell carcinoma. Methods: MicroRNAs differentially expressed in bladder and adjacent tissues were screened by Mi-croRNA. The expression of miR-200c and miR-141 in bladder cancer and bladder cancer cell lines was verified by real-time RT-PCR. Results: The expression intensity of MiR-200c and miR-141 in bladder tumor tissue was higher than that in adjacent normal bladder mucosa, and in superficial bladder tumor was higher than that in muscle-invasive invasive bladder tumor. In the high-grade group, the bladder tumor was higher than the low-grade group , Which was highly expressed in well-differentiated and less malignant 5637 cell lines and lower in poorly differentiated and malignant T24, J82 and EJ cell lines. Conclusion: MiR-200c and miR-141 may be involved in the progression of bladder cancer.
