目的通过比较不同临床分期大肠癌组织的差异表达蛋白,以发现与大肠癌临床分期相关蛋白。方法将不同临床分期大肠癌组织蛋白进行二维凝胶电泳,选择部分差异表达蛋白进行MALD I-TOF质谱分析和生物信息学分析。结果建立了不同临床分期大肠癌组织的二维凝胶电泳图谱,其中Ⅰ期,Ⅱ期,Ⅲ期,Ⅳ期大肠癌组织平均蛋白质点数分别为970±41、980±32、1010±43、1240±34;以Ⅰ期大肠癌为参照,Ⅱ期大肠癌差异表达蛋白有52.00±12,Ⅲ期大肠癌差异表达蛋白42.00±11,Ⅳ期大肠癌差异表达蛋白72.00±15。通过进行质谱分析和生物信息学查询,鉴定了显著差异表达的19个蛋白点,其中Ⅱ期,Ⅲ期,Ⅳ期均上调的有3个蛋白,而仅在Ⅳ期中上调蛋白有1个。结论不同临床分期的大肠癌中存在着差异表达蛋白,这些蛋白可能是大肠癌诊断和预后的标志物。
OBJECTIVE: To compare differentially expressed proteins in different clinical stages of colorectal cancer and to identify the proteins involved in the clinical staging of colorectal cancer. Methods Two-dimensional gel electrophoresis of different clinical staging of colorectal cancer tissue proteins was carried out, and some differentially expressed proteins were selected for MALDI-TOF mass spectrometry and bioinformatics analysis. Results The two-dimensional gel electrophoresis patterns of colorectal cancer with different clinical stages were established. The average protein spots of stage Ⅰ, stage Ⅱ, stage Ⅲ and stage Ⅳ colorectal cancer tissues were 970 ± 41,980 ± 32,1010 ± 43,1240 ± 34; differentially expressed protein of stage Ⅱ colorectal cancer was 52.00 ± 12, stage Ⅲ colorectal cancer differentially expressed protein 42.00 ± 11, stage Ⅳ colorectal cancer differentially expressed protein 72.00 ± 15. Nineteen protein spots that were significantly differentially expressed were identified by mass spectrometry and bioinformatics queries, of which three were up-regulated in stages II, III, and IV, and one was up-regulated only in stage IV. Conclusion There are differentially expressed proteins in different clinical stages of colorectal cancer. These proteins may be the markers of colorectal cancer diagnosis and prognosis.
