目的:建立测定蛤蚧颗粒中黄芪甲苷的HPLC-ELSD法。方法:色谱柱为Phenomenex C18(4.60mm×250mm,5μm),流动相是乙腈-水(32:68),进样量20μL,流速1.0mL/min,柱温30℃,漂移管温度90℃,氮气流量2.75L/min。结果:回归方程线性关系良好,回归方程为Y=1.5307X+5.6229,r=0.9998,n=5,线性范围为2.03~10.14μg,加样回收率为99.2%,其RSD为1.09%。结论:该方法适用于测定蛤蚧颗粒中黄芪甲苷的含量,结果准确可靠,重现性好,能有效地控制蛤蚧颗粒的质量。
Objective: To establish a HPLC-ELSD method for the determination of astragaloside IV in loquat granules. Methods: The chromatographic column was Phenomenex C18 (4.60mm×250mm, 5μm), the mobile phase was acetonitrile-water (32:68), the injection volume was 20μL, the flow rate was 1.0mL/min, the column temperature was 30°C, and the drift tube temperature was 90°C. Nitrogen flow rate is 2.75L/min. Results: The regression equation has a good linear relationship, the regression equation is Y=1.5307X+5.6229, r=0.9998, n=5, and the linear range is 2.03~10.14μg. The recovery rate is 99.2%, and the RSD is 1.09%. Conclusion: This method is suitable for the determination of astragaloside IV in loquat granules. The results are accurate, reliable, and reproducible, and can effectively control the quality of wolfberry granules.
