目的:研究连翘酯苷对PC12细胞增殖及其损伤模型的影响。方法:应用谷氨酸(20 mmol·L-1)、低糖低血清培养基及β淀粉样蛋白25~35(Aβ25~35,10μmol·L-1)建立谷氨酸、低糖低血清及Aβ25~35致PC12细胞损伤模型,观察连翘酯苷低、中、高浓度(0.1,1,5μmol·L-1)剂量对上述模型细胞存活率的影响,同时观察连翘酯苷低、中剂量对Aβ25~35致PC12细胞凋亡的影响。结果:连翘酯苷可提高PC12细胞存活率,提高谷氨酸、低糖低血清及Aβ25~35致PC12细胞损伤模型的存活率,与模型组比较,有显著性差异(P<0.05),并呈现一定量效关系。连翘酯苷(0.1,1μmol·L-1)可降低Aβ25~35引起PC12细胞凋亡率(P<0.05,P<0.01)。结论:连翘酯苷在体外实验具有神经细胞保护作用。
Objective: To study the effects of forsythiaside on the proliferation and injury of PC12 cells. Methods: Glutamic acid, low-glycemic serum and Aβ25-β were determined by glutamic acid (20 mmol·L -1), low glucose and low serum medium, and amyloid β 25-35 (Aβ 25-35 and 10 μmol·L -1) 35 induced PC12 cell injury model to observe the effects of Forsythiaside at low, medium and high concentrations (0.1, 1, 5μmol·L-1) on the cell viability of the above models, and observe the effects of Forsythiaside at low and medium doses Effect of Aβ25 ~ 35 on Apoptosis of PC12 Cells. Showing a certain amount of effective relationship. Forsythiaside (0.1,1μmol·L-1) decreased the apoptosis rate of PC12 cells induced by Aβ25-35 (P <0.05, P <0.01). Conclusion: Forsythiaside has neuroprotective effect in vitro.
