目的通过对人类重组型和野生型酸性成纤维细胞生长因子(raFGF,waFGF)的比较研究,探讨raFGF与促分裂性有关的生物学作用。方法以NIH3T3细胞株,采用MTT法通过细胞增殖实验检测waFGF和raFGF的促分裂性,通过流式细胞术检测细胞凋亡、细胞膜通透性和线粒体膜电位;同时,检测肝素(HS)对aFGF的上述作用的影响。结果raFGF对细胞增殖作用明显低于waFGF,对细胞凋亡的抑制作用及其对细胞膜通透性的增强作用也明显低于waFGF,对线粒体膜电位的增强作用明显低于waFGF。HS使aFGF的上述作用增强。结论raFGF的促分裂作用低于waFGF,而对细胞凋亡的影响不明显;HS对aFGF促分裂作用具有促进效应。
OBJECTIVE: To investigate the biological role of raFGF in mitogenicity by comparing human recombinant and wild-type acidic fibroblast growth factor (sFGF, waFGF). Methods NIH3T3 cell line was used to detect the mitogenic activity of waFGF and raFGF by MTT assay. Cell apoptosis, cell membrane permeability and mitochondrial membrane potential were measured by flow cytometry. Meanwhile, the effect of heparin on aFGF The impact of the above effects. Results The effect of raFGF on cell proliferation was significantly lower than that of waFGF. The inhibitory effect on cell apoptosis and the enhancement of cell membrane permeability of raFGF were also significantly lower than those of waFGF. The enhancement of mitochondrial membrane potential was significantly lower than that of waFGF. HS enhances the above effects of aFGF. Conclusion The mitogenic effect of raFGF is lower than that of waFGF, but the effect of raFGF on apoptosis is not obvious. HS can promote the mitosis of aFGF.
