目的:建立测定天麻药材中游离天麻素和总天麻素含量的方法。方法:采用药典方法测定游离天麻素的含量;用碱水解-HPLC法,测定总天麻素的含量。色谱条件:采用LUNA C18(2)(4.6 mm×150 mm,5μm)色谱柱,流动相为乙腈-0.05%磷酸(3∶97),流速0.8 mL.min-1,检测波长220 nm。结果:天麻素进样量在0.097～0.484μg范围内有良好线性关系(r=0.9997),平均回收率(n=6)为99.0%。结论:该方法简便,可行,重现性好,可用于控制天麻药材质量。
Objective: To establish a method for determination of free gastrodin and total gastrodin in Gastrodia elata Blume. Methods: The content of free gastrodin was determined by pharmacopoeia method. The content of total gastrodin was determined by alkaline hydrolysis-HPLC method. Chromatographic conditions: The mobile phase consisted of acetonitrile-0.05% phosphoric acid (3:97) with a flow rate of 0.8 mL.min-1 at a wavelength of 220 nm using a LUNA C18 (2) (4.6 mm × 150 mm, 5 μm) column. Results: Gastrodin injection had good linearity (r = 0.9997) in the range of 0.097 ~ 0.484μg with the average recovery of 99.0% (n = 6). Conclusion: The method is simple, feasible, reproducible, and can be used to control the quality of Gastrodia elata Blume.
