[目的]为找到适合蝇类生物防治的病原微生物,对罹病棕尾别麻蝇Boettcherisca peregrine体上的病原物进行分离纯化。[方法]利用光学显微镜镜检菌丝和分生孢子的形态,根据病原菌的培养性状、菌丝和分生孢子的形态进行鉴定,并对其生物学特性和致病性进行初步研究。[结果]棕尾别麻蝇体上的病原物为蜡蚧轮枝菌Verticillium lecanii,将该菌株编号为KMZW-1。该菌株菌落生长最适培养基为PDA培养基,最适温度为29℃,最适pH值为6.0。其孢子悬浮液对棕尾别麻蝇、丝光绿蝇、家蝇、酪蝇、黑腹果蝇成虫的致死中浓度(LC50)分别为9.50×105、4.58×107、4.06×107、4.10×103和1.05×107分生孢子/ml,致死中时间(LT50)分别为6.86、8.17、8.16、8.12和3.22d。[结论]蜡蚧轮枝菌KMZW-1是一种对5种蝇类成虫具有活性的病原菌。
[Objective] The study aimed to isolate and purify pathogenic microorganisms from the diseased Boettcherisca peregrine of diseased mantis to find pathogenic microorganisms suitable for the biological control of flies. [Method] The morphology of mycelium and conidia were observed by light microscopy. The pathogen was identified according to the culture traits, mycelium and conidia morphology, and its biological characteristics and pathogenicity were studied. [Result] The pathogen of Verticillium lecanii was named as KMZW-1. The optimal culture medium for this strain was PDA medium, the optimum temperature was 29 ℃ and the optimum pH was 6.0. The lethal concentration (LC50) of its spore suspension against adult Bemisia matsudana, Meriones unguiculatus, Musca domestica, Chestnut and Drosophila melanogaster were 9.50 × 105,4.58 × 107,4.06 × 107,4.10 × 103 and 1.05 × 107 conidia / ml and lethal time (LT50) were 6.86,8.17,8.16,8.12 and 3.22d, respectively. [Conclusion] Verticillium lecanii KMZW-1 is a pathogen that is active against five species of flies.
