目的评价鹿角脱盘5种提取部位对肝癌H_(22)荷瘤小鼠免疫功能的影响。方法将90只SPF级昆明种小鼠随机分成正常阴性对照组、模型组、阳性对照组、鹿角脱盘酸(盐酸)、碱(氢氧化钠)、盐(氯化钠)、醇(乙醇)和水5种提取部位组和鹿角脱盘原粉组共9组,每组10只。分别以脾脏组织病理变化、外周血和脾脏CD4~+和CD8~+T淋巴细胞百分比、血清中细胞因子IL-6、IL-12和抗体IgE、IgG、IgM水平为指标,评价鹿角脱盘的盐酸、氢氧化钠、氯化钠、乙醇和水5种提取部位提高荷瘤小鼠免疫功能的作用。结果与模型组比较,鹿角脱盘5种提取部位能够保护荷瘤小鼠脾脏结构;提高脾脏和外周血CD4~+/CD8~+比值(P<0.05);提高荷瘤小鼠血清中细胞因子IL-6、IL-12和抗体IgE、IgG、IgM的水平(P<0.05)。结论鹿角脱盘碱溶液提取部位效果最好;鹿角脱盘可能通过提高荷瘤小鼠体液免疫和细胞免疫的功能,逆转肿瘤发生发展对机体免疫功能的抑制作用,发挥抗肿瘤作用。
Objective To evaluate the effect of antler extraction on the immune function of H22 tumor-bearing mice with hepatocellular carcinoma. Methods Ninety SPF Kunming mice were randomly divided into normal negative control group, model group, positive control group, antler acid (hydrochloric acid), alkali (sodium hydroxide), salt (sodium chloride), alcohol (ethanol) Water and five kinds of extraction site group and antler off the original powder group of 9 groups, 10 in each group. The pathological changes of spleen, the percentage of CD4 ~ + and CD8 ~ + T lymphocytes in peripheral blood and spleen, the levels of IL-6, IL-12 and IgE, IgG and IgM in serum Hydrochloric acid, sodium hydroxide, sodium chloride, ethanol and water extract of 5 parts enhance the immune function in mice. Results Compared with the model group, the five kinds of extraction sites of antler offspring could protect the spleen of tumor-bearing mice; increase the ratio of CD4 ~ + / CD8 ~ + in spleen and peripheral blood (P <0.05); increase the serum levels of cytokines IL-6, IL-12 and IgE, IgG, IgM levels (P <0.05). Conclusion Antler detached from the alkaline solution has the best effect. The removal of antler may exert anti-tumor effect by reversing the inhibitory effect of tumor development and development on the immune function of the tumor-bearing mice by increasing humoral and cellular immunity.
