目的 :探讨力尔凡对舌癌细胞的抑制作用及作用机制。方法 :利用MTT方法检测力尔凡对舌癌细胞的抑制作用 ,末端脱氧核苷酰转移酶 (TUNEL法 )标记免疫荧光染色法检测细胞周期 ,细胞免疫化学方法 (SPTM)分析p53基因表达。结果 :力尔凡对舌癌细胞抑制作用的IC50为342.83μg/ml,力尔凡对舌癌细胞周期具有调控作用 ,浓度为100μg/ml时 ,G0~G1 期为59.02 % ,G2-M期为19.59% ,S期无明显改变 ;细胞免疫化学分析结果发现力尔凡具有上调舌癌细胞 p53基因表达作用。结论 :力尔凡具有抑制舌癌细胞增殖作用 ,作用机理是通过上调 p53基因表达使细胞周期阻断于G1 期。
Objective: To investigate the inhibitory effect and its mechanism of Li Er Fan on tongue cancer cells. Methods: MTT assay was used to detect the inhibitory effect of Riehlmann on tongue cancer cells. The cell cycle was detected by TUNEL method and the expression of p53 gene was analyzed by immunocytochemistry (SPTM). Results: The average inhibitory effect of Rievan on tongue cancer cells was 342.83μg / ml, and Rievan had regulative effect on the cell cycle of tongue cancer cells. The G0-G1 phase was 59.02% at the concentration of 100μg / ml, G2-M phase 19.59%, S phase had no significant change; cellular immunochemical analysis of the results found that Rievan has the role of upregulation of p53 gene expression in tongue cancer cells. Conclusion: Rievan can inhibit the proliferation of tongue cancer cells. The mechanism is that the cell cycle is blocked in G1 phase by up-regulating p53 gene expression.
