目的:探讨妊娠子宫自然杀伤细胞(uterinenaturalkillercell,uNK)和外周血NK细胞(peripheralnaturalkillercell,pNK)中乏氧诱导因子(hypoxiainduciblefactor,HIF)和血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)C、VEGFE的表达。方法:采用免疫磁珠法分离纯化妊娠早期uNK和pNK细胞,提取其总RNA,采用RT-PCR技术检测HIF和VEGF的mRNA在人妊娠早期uNK细胞和pNK细胞中的表达。结果:①人妊娠早期uNK细胞中HIFmRNA表达强于pNK细胞(2.31±0.19和0.12±0.02);②VEGFC和VEGFE在两种细胞中均有表达,但VEGFE在uNK中的表达强于pNK(0.82±0.03和0.10±0.01),VEGFC在两种细胞中的表达相近。结论:人妊娠早期uNK细胞HIF和VEGFE的强表达可能与uNK细胞在胎盘血管形成中的作用密切相关。
Objective: To investigate the expression of hypoxia induced factor (HIF) and vascular endothelial growth factor (VEGF) C, VEGFE in uterine natural killer cells (uNK) and peripheral natural killer cells (pNK) in pregnant women. Methods: The uNK and pNK cells were isolated and purified by immunomagnetic beads method. The total RNA was extracted and the expression of HIF and VEGF mRNA was detected by RT-PCR in human uNK and pNK cells. Results: ① The expression of HIF mRNA in uNK cells was higher than that in pNK cells in early pregnancy (2.31 ± 0.19 and 0.12 ± 0.02). ② The expression of VEGF and VEGFE in uNK cells was stronger than that in pNK cells (0.82 ± 0.03 and 0.10 ± 0.01). The expression of VEGFC in both cells was similar. Conclusion: The strong expression of HIF and VEGFE in uNK cells in early pregnancy may be closely related to the role of uNK in placental angiogenesis.
