复健片对MCAO大鼠脑组织GFAP表达及局部脑血流量的影响

目的:探讨复健片对MCAO模型大鼠星形细胞活性及其局部脑血流灌注状态的影响。方法:选取健康雄性Wistar大鼠,随机分为3组,即假手术组、模型对照组(简称模型组)、药物组各24只。采用右侧近端大脑中动脉电凝术,制作大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)模型。按要求分别给予复健片水溶液、生理盐水灌胃2周。采用激光多普勒血流仪测定梗死侧额叶局部脑血流量(regional cerebral blood flow,rCBF);利用免疫组织化学方法,观察复健片对MCAO大鼠脑组织胶质纤维酸性蛋白(glial fibrillary acid protein,GFAP)表达的影响。指标间的相关性采用直线相关分析。结果:rCBF测量结果表明,术后模型组与药物组梗死侧额叶皮质rCBF均显著低于假手术组(P<0.01);而经药物治疗1周后,梗死侧额叶皮质的rCBF虽然仍低于假手术组水平(P<0.05),但较之治疗前和同期模型组,均有明显增加(P<0.05);经药物治疗2周后,梗死侧额叶皮质的rCBF仍高于同期模型组(P<0.05);模型组与假手术组在观察的各时点则均无显著性变化。免疫组化结果显示,大脑中动脉阻塞后,模型大鼠梗死灶周围区GFAP的表达同假手术组相比并无显著改变(P>0.05);而药物组分别给药1周、2周后,GFAP的表达均较同期模型组、假手术组及药物组术后1周有显著增强(P<0.05)。药物灌胃1周、2周组的GFAP平均灰度值和rCBF相关分析表明,两者具有明显相关性。结论:复健片可以增强MCAO模型大鼠脑组织星形胶质细胞的活性,而且这种活性的上调有助于改善受损脑组织的局部血流量。 Objective: To investigate the effect of Fufang Tablet on the activity of astrocytes and the local cerebral perfusion in MCAO rats. Methods: Healthy male Wistar rats were randomly divided into three groups: sham operation group, model control group (model group) and drug group (24 rats each). Middle cerebral artery occlusion (MCAO) model was made by right proximal middle cerebral artery electrocoagulation. According to the requirements of rehabilitation tablets were given aqueous solution, saline for 2 weeks. The regional cerebral blood flow (rCBF) of the frontal lobe was measured by laser Doppler flowmetry. The expression of glial fibrillary acid protein, GFAP) expression. Results: The results of rCBF measurement showed that the rCBF of the frontal cortex in the model group and the drug group was significantly lower than that in the sham operation group (P <0.01). After 1 week of treatment, the rCBF in the infarct side frontal cortex (P <0.05), but significantly increased compared with the model group before and after treatment (P <0.05). After 2 weeks of treatment, the rCBF in the infarct side frontal cortex was still higher than that in the sham-operation group Model group (P <0.05). There was no significant change in the model group and the sham operation group at each time point. The results of immunohistochemistry showed that there was no significant difference in the expression of GFAP in the peri-infarct area between the model group and the sham operation group (P> 0.05) after occlusion of the middle cerebral artery, while the drug group was administered for 1 week and 2 weeks respectively , GFAP expression than the same period model group, sham operation group and drug group 1 week after surgery was significantly enhanced (P <0.05). The GFAP mean gray value and rCBF correlation analysis showed that there was a significant correlation between the two. Conclusion: The tablets can enhance the activity of astrocytes in brain tissue of MCAO rats, and the upregulation of this activity can help to improve the local blood flow in damaged brain tissue.
