建立整体化色谱柱分离分析甘草酸18H-差向异构体的方法。采用C18整体化色谱柱,流动相为磷酸盐缓冲液-乙腈体系,探讨了磷酸盐缓冲液pH及浓度、乙腈含量、柱温等对甘草酸18H-差向异构体分离的影响,在优化色谱条件下,18α-、18β-甘草酸得到基线分离,分离度可达2.75,二异构体出峰时间在10 min内,且在2.12~21.2μg/mL范围内线性关系良好。运用整体化色谱柱缩短了甘草酸18H-差向异构体的分析时间,提高了分析效率,方法可运用于评价甘草酸的异构化及质量控制。
To establish a method of separation and analysis of 18H-epimer of glycyrrhizin by using the integrated column. The effect of 18H-epimer on the separation of glycyrrhizin 18H was studied by using C18 monolithic column and mobile phase of phosphate buffer-acetonitrile system. The effects of pH and concentration of phosphate buffer, acetonitrile and column temperature on the separation of 18H- Under the chromatographic conditions, 18α-, 18β-glycyrrhizinate was separated baseline with a resolution of 2.75, peak time of the two isomers within 10 min, and good linearity in the range of 2.12-21.2μg / mL. The use of monolithic columns reduces the analysis time of glycyrrhizin 18H-epimer and improves the analytical efficiency. The method can be applied to evaluate the isomerization and quality control of glycyrrhizin.
