对安徽省长丰县随机采集的20个草莓根腐病病样进行病原鉴定,结果分离获得12个菌株。根据培养特征和形态学鉴定,确定分离到的菌株均为尖孢镰刀菌(Fusarium oxysporum);选取真菌通用引物ITS1/ITS4对菌株基因组DNA进行PCR扩增并进行克隆和测序,通过与NCBI数据库序列比对,分子鉴定结果与形态学一致。将分离到的菌株回接到健康草莓幼苗上,2~3周后植株表现出与田间发病相似的症状,重新分离能够得到相同的病原物。基于形态学、分子生物学鉴定及致病性检测,最终确定安徽省长丰县草莓根腐病的病原菌为Fusarium oxysporum。
Pathogen identification of 20 strawberry root rot samples randomly collected in Changfeng County of Anhui Province was carried out. As a result, 12 strains were isolated. The isolates were identified as Fusarium oxysporum according to their culture characteristics and morphological characteristics. The common genomic DNA of ITS1 / ITS4 was used to amplify and clone the genomic DNA of the isolates. After cloning and sequencing with the NCBI database, Comparison, molecular identification and morphological findings. The isolated strains were returned to healthy strawberry seedlings and the plants showed symptoms similar to the onset of disease in the field in 2 to 3 weeks and the same pathogen was recovered by re-isolation. Based on morphological, molecular biological identification and pathogenicity test, the pathogen of strawberry root rot was identified as Fusarium oxysporum in Changfeng County of Anhui Province.
