丹瓜方对ApoE-/-糖尿病模型小鼠脑组织Caspase-3蛋白及Bcl-2、Bax mRNA表达水平的影响

目的 研究丹瓜方对糖尿病模型Apo E-/-小鼠糖脂代谢、脑组织胱冬肽酶-3(Caspase-3)蛋白表达及β淋巴细胞瘤基因-2(Bcl-2)、Bax mRNA表达水平的影响.方法 采用链脲佐菌素(streptozo tocin,STZ)注射法对Apo E-/-小鼠制备糖尿病模型.将成模后的32只小鼠按体重分层,再按血糖水平排序,采用随机数字表分为模型组[无菌水(SW),15 mL/(kg·d)]、丹瓜方组[丹瓜方(DGR),15 mL/(kg· d)]、联合组[吡格列酮(PG) 4.3 mg/(kg·d) +DGR 15 mL/(kg·d)]及吡格列酮组[PG,4.3 mg/(kg·d)],每组8只.另选血糖正常的8只同龄同品系C57小鼠作为对照组.灌胃干预12周.监测空腹血糖(fasting blood glucose,FBG),测定血清TC、LDL-C水平;实时荧光定量PCR(qPCR)法检测ApoE-/-小鼠脑组织Bcl-2、Bax mRNA表达,采用Western blot法测定脑组织Caspase-3蛋白表达水平.结果 与对照组比较,模型组Apo E-/-小鼠血清FBG、TC、LDL-C及脑组织Caspase-3表达水平升高(P<0.01);与模型组比较,丹瓜方组FBG、TC及LDL-C水平降低,Bcl-2、Bax mRNA水平升高(P<0.01,P<0.05).丹瓜方组Caspase-3蛋白表达水平低于模型组、联合组及吡格列酮组各组(P<0.01);联合组Bcl-2/Bax比值高于模型组(P<0.05),丹瓜方组高于吡格列酮组(P<0.01).结论 丹瓜方可调节糖尿病糖脂代谢及脑组织Caspase-3蛋白及Bcl-2、Bax mRNA表达,可能对糖尿病造成的脑组织细胞凋亡有一定的防治作用.“,”Objective To observe the effects of Dangua Recipe (DGR) on glycolipid metabolism,Caspase-3 protein expression,and mRNA expression levels of Bcl-2 and Bax in brain tissue of transgenic ApoE-/-model mice.Methods Diabetes model was prepared by peritoneally injecting streptozotocin (S-rz) to ApoE-/-mice.Totally 32 successfully modeled mice were stratified by body weight,and then sorted by blood glucose level from low to high.They were randomly divided into the model group [MOD,fed with sterile water (SW),at the daily dose of 15 mL/kg],the DGR group (at the daily dose of 15 mL/kg),the combination group [COM (fed with DGR at the daily dose of 15 mL/kg) and pioglitazone (at the daily dose of 4.3 mg/kg)],and the pioglitazone group (PG,at the daily dose of 4.3 mg/kg) according to random number table,8 in each group.Besides,another 8 euglycemia C57 mice of the same age and strain were recruited as the control group.The intervention lasted for 12 weeks by gastrogavage.Fasting blood glucose (FBG) was monitored.Levels of total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) were detected as well.mRNA expressions of Bcl-2 and Bax in brain tissue were detected by Real-time fluorescent quantitative PCR (qPCR).Caspase-3 protein expression level in brain tissue was detected by Western blot.Results Levels of serum FBG,TC,LDL-C,and Caspase-3 expression in brain tissue were higher in the MOD group than in the control group (P <0.01).Levels of FBG,TC,LDL-C were lower in the DGR group than in the MOD group,while mRNA expressions of Bcl-2 and Bax were higher in the DGR group than in the MOD group (P <0.01,P <0.05).Caspase-3 expression level was lower in the DGR group than in the other groups (P <0.01).Bcl-2/Bax ratio was higher in the COM group than the MOD group (P <0.05).Bcl-2/Bax ratio was higher in the DGR group than the PG group (P <0.01).Conclusions DGR could regulate glycolipid metabolism,expressions of Caspase-3protein and Bcl-2/Bax mRNA.It might possibly play effective roles in preventing and treating diabetes induced apoptosis brain tissue cells.
