目的观察抗纤灵对5/6肾切除小鼠肾组织转化生长因子-β(TGF-β)、结缔组织生长因子(CTGF)表达的影响。方法随机从50只C57小鼠中取40只建立5/6肾切除慢性肾衰肾纤维化模型,术后2周根据肌酐值将40只小鼠分为抗纤灵低、中、高剂量治疗组和模型组,每组10只。另10只小鼠为假手术组。治疗12周后,用western blot法、RT-q PCR法分别测定各组小鼠肾组织TGF-β、CTGF蛋白及mRNA表达水平。结果模型组小鼠肾组织TGF-β、CTGF蛋白及mRNA表达较假手术组升高(P<0.01);与模型组比较,抗纤灵中、高剂量组小鼠肾组织TGF-β及CTGF蛋白、mRNA表达下调(P<0.01),两剂量组间TGF-β、CTGF的蛋白及mRNA表达差异无统计学意义(P>0.05)。结论抗纤灵中、高剂量治疗组均能下调5/6肾切除小鼠肾组织TGF-β、CTGF蛋白及mRNA表达,两剂量治疗组间尚不存在量效相关性。提示抗纤灵可能通过调控TGF-β及其下游因子延缓肾纤维化,保护残肾功能。
Objective To observe the effect of Kangxianling on the expression of transforming growth factor-β (TGF-β) and connective tissue growth factor (CTGF) in 5/6 nephrectomized mice. Methods A total of 40 C57 mice were randomly divided into 5 groups of 5 rats with chronic kidney failure and 5 weeks of nephrectomy. According to the creatinine value, 40 mice were divided into low, middle and high doses Group and model group, 10 rats in each group. The other 10 mice were sham operation group. After 12 weeks of treatment, the expression of TGF-β and CTGF protein and mRNA in renal tissue of each group were determined by western blot and RT-q PCR respectively. Results The expression of TGF-β and CTGF protein and mRNA in renal tissue in model group were significantly higher than those in sham-operated group (P <0.01). Compared with model group, the expression of TGF-β and CTGF (P <0.01). There was no significant difference in the expression of TGF-β and CTGF between the two groups (P> 0.05). Conclusion Both Kangxiling and high-dose treatment groups can down-regulate the expression of TGF-β and CTGF protein and mRNA in renal tissues of 5/6 nephrectomized mice. No dose-response relationship exists between the two doses of treatment group. Tip Kangxiao Ling may regulate TGF-β and its downstream factors delay renal fibrosis, residual renal function protection.
