目的:探讨乙型病毒性肝炎(HBV)患者组织中病毒抗原与 Fas 抗原表达的关系。方法:采用免疫组化双标记技术,以鼠抗-HBs 单克隆抗体、兔抗-HBc 及免抗-Fas 多克隆抗体,对急性普通型、慢性活动型肝炎及活动性肝硬化各20例肝组织中 HBVAg 及 Fas 抗原的表达及分布进行了检测和比较。结果:60例肝组织中 HBsAg、HBcAg 和 Fas 抗原的检出率分别为81.6%(49/60)、43.3%(26/60)及76.7%(46/60),三种不同的组织学类型中 HBVAg 和 Fas 抗原的检出率均无显著差别(X~2=2.93及2.41;P 值>0.25)。肝组织中 Fas 抗原均于肝细胞浆内表达,在急性普通型肝炎,阳性细胞在小叶内弥散分布;慢性活动型肝炎和活动性肝硬化则更多集聚在小叶或假小叶周边及碎屑样坏死灶与单个核细胞毗邻处。免疫组化双标记染色显示病毒抗原及 Fas 抗原可在同一细胞或同一区域不同细胞内表达,也可在不同区域表达,Fas 抗原阳性细胞明显多于病毒抗原性细胞,而正常的肝组织内无 Fas 抗原表达。结论:病毒性肝炎时,肝细胞内的病毒抗原表达可以诱导Fas 抗原的表达,并与肝细胞的凋亡有关。
Objective: To investigate the relationship between viral antigen and Fas antigen in tissues of patients with viral hepatitis B (HBV). Methods: The anti-HBs monoclonal antibody, rabbit anti-HBc and anti-Fas polyclonal antibody were used in immunohistochemical double-labeling technique to detect the expression of 20 common hepatic fibrosis cells in acute common, chronic active hepatitis and active cirrhosis The expression and distribution of HBVAg and Fas antigen in tissues were detected and compared.
