[目的]了解草鱼干扰素调节因子(IRF)的功能,为研究鱼类天然免疫系统提供理论基础。[方法]利用RTPCR技术检测草鱼(Ctenopharyngodon idella)IRF4a、IRF4b、IRF4-like、IRF8和IRF9的早期胚胎表达、组织表达以及草鱼呼肠孤病毒(grass carp reovirus,GCRV)体内诱导表达。[结果]胚胎表达结果显示IRF4a、IRF8和IRF9分别显著表达于0 hpf、0 hpf和30 hpf(P<0.05),IRF4b、IRF4-like表达变化不明显;组织表达结果显示IRF4a、IRF4b、IRF9在鳃中表达最高,IRF4-like、IRF8在脾脏中表达最高。IRF4a、IRF4b和IRF9在感染4 h后表达量显著上调(P<0.05),IRF4-like在感染1 h和24 h时表达量显著上调(P<0.05),IRF8在感染48 h后表达量显著上调(P<0.05)。[结论]胚胎期IRF4a、IRF8存在母源表达现象,能够保护胚胎免受外界干扰,直至草鱼免疫系统完善。IRF4亚家族成员在免疫器官鳃和脾脏中高表达。IRF4a、IRF4b、IRF9和IRF4-like在GCRV病毒感染的前期和中前期发挥作用,而IRF8主要在感染的中后期发挥抗病毒的作用。
[Objective] To understand the function of grass carp interferon regulatory factor (IRF) and provide the theoretical basis for studying the natural immune system of fish. [Method] The early embryo expression, tissue expression and inducible expression of grass carp reovirus (GCRV) in IRF4a, IRF4b, IRF4b, IRF4-like, IRF8 and IRF9 of grass carp [Results] The results of embryo expression showed that IRF4a, IRF8 and IRF9 were significantly expressed at 0 hpf, 0 hpf and 30 hpf respectively (P <0.05), while the expressions of IRF4b and IRF4-like did not change significantly. The expression of IRF4a, IRF4b and IRF9 Gill highest expression, IRF4-like, IRF8 highest expression in the spleen. The expression of IRF4a, IRF4b and IRF9 were significantly up-regulated 4 h after infection (P <0.05). The expression of IRF4-like was significantly up-regulated at 1 h and 24 h (P <0.05) (P <0.05). [Conclusion] The maternal expression of IRF4a and IRF8 in embryo could protect embryos from external disturbance until the immune system of grass carp was perfect. IRF4 subfamily members are highly expressed in gill and spleen of immune organs. IRF4a, IRF4b, IRF9 and IRF4-like play an important role in the early and middle stages of GCRV infection, whereas IRF8 plays an antiviral role mainly in the mid- and late-stage infections.
