目的 :研究大豆异黄酮对去卵巢大鼠骨密度以及雌激素活性的影响。方法 :将 10~ 12月龄的雌性Wistar大鼠随机分为 6组 :假手术组 (SHAM )、切卵巢模型组 (OVX)、尼尔雌醇组 (OVX E)、小剂量异黄酮组 (L ISO)、中剂量异黄酮组 (M ISO)、大剂量异黄酮组 (H ISO) ,每组 8只。后 5组大鼠被切除双侧卵巢 ,SHAM组只被切除卵巢附近脂肪组织。L ISO ,M ISO ,H ISO分别灌胃给予 30 ,6 0 ,12 0mg·kg- 1的大豆异黄酮 ,OVX E组大鼠灌胃给予0 .2mg·kg- 1·wk- 1的尼尔雌醇 ,SHAM与OVX组以等剂量的溶剂灌胃 ,15wk后股动脉放血处死动物 ,收集血液用于血清碱性磷酸酶活性、血清雌二醇水平、血钙、血磷测定 ,分离出右侧股骨、第 2腰椎用于骨密度测量 ,测定双侧子宫重量。结果 :与SHAM组相比 ,OVX组股骨骨密度和椎骨骨密度均可见不同程度降低 (P <0 .0 5和P >0 .0 5 ) ,尼尔雌醇与异黄酮 12 0mg·kg- 1能明显升高去卵巢大鼠的股骨和椎骨骨密度 (P <0 .0 5 )。OVX组的血清碱性磷酸酶水平高于SHAM组 (P <0 .0 5 ) ,异黄酮6 0mg·kg- 1可降低去卵巢大鼠的血清碱性磷酸酶水平 (P <0 .0 5 )。OVX组的血清雌二醇水平与子宫系数明显低于SHAM组 (P <0 .0 1) ,异黄酮的不同剂量组与OVX E组的血清雌二醇水平、子宫系数都明显高于OV
Objective: To study the effect of soy isoflavones on bone mineral density and estrogen activity in ovariectomized rats. Methods: Female Wistar rats (10-12 months old) were randomly divided into 6 groups: sham operation group (OVX), ovariectomized group (OVX E), low dose isoflavone group L ISO), medium dose isoflavone group (M ISO) and high dose isoflavone group (H ISO), with 8 in each group. After 5 rats were removed bilateral ovaries, SHAM group was only excised near the ovary adipose tissue. L ISO, M ISO and H ISO were intragastrically administered with 30, 60 and 120 mg · kg -1 of soy isoflavones respectively. OVX E rats were given intragastric administration of 0.2 mg · kg -1 · wk -1 of Neal In the estradiol, SHAM and OVX groups, the rats were orally administered with the same dose of solvent. After 15 weeks, the femoral artery was sacrificed and the blood collected for determination of serum alkaline phosphatase, serum estradiol, serum calcium and phosphorus, and the right Side femur, the second lumbar spine for bone mineral density measurement, determination of bilateral uterine weight. Results: Compared with SHAM group, the femoral BMD and vertebral BMD of OVX group were all decreased to some extent (P <0.05 and P> 0.05), while the nalteral sterol and isoflavone 120 mg · kg- 1 significantly increased femur and vertebral bone mineral density in ovariectomized rats (P <0.05). Serum alkaline phosphatase level in OVX group was higher than that in SHAM group (P <0.05), and isoflavone 60 mg · kg -1 could decrease serum alkaline phosphatase level in ovariectomized rats (P <0.05) ). Serum estradiol level and uterine coefficient in OVX group were significantly lower than those in SHAM group (P <0.01). Serum estradiol level and uterine coefficient in different dose groups of isoflavone and OVX E group were significantly higher than OV
