目的探讨经股静脉移植的大鼠真皮多能干细胞在缺血性损伤脑组织中的存活、迁移及分布情况。方法分离培养人真皮来源的多能干细胞(skin-derived precursors,SKPs),采用线栓法制作大鼠局灶性脑缺血再灌注模型,缺血2h后行再灌注,于再灌注24h后将CM-DiI标记的人真皮来源的SKPs经大鼠股静脉输入动物模型体内,植入后分别在第1、3、5d处死大鼠,作冰冻切片,采用免疫组织荧光染色法,检测SKPs在脑缺血大鼠体内的存活、迁移及分布情况。结果在移植了SKPs后的MCAO大鼠脑组织冰冻切片中,荧光显微镜下可见移植的SKPs。静脉移植的细胞主要分布在缺血灶的周围,健侧脑组织中偶见移植细胞。结论经股静脉移植的SKPs,能够在宿主缺血的脑组织中存活,并向损伤部位迁移。
Objective To investigate the survival, migration and distribution of rat dermal multipotent stem cells transplanted with femoral vein in ischemic brain tissue. Methods Human dermal dermal derived stem cells (SKPs) were isolated and cultured. The focal cerebral ischemia / reperfusion model was established by thread occlusion. After 2 hours of ischemia, reperfusion was performed. After 24 hours of reperfusion, Human dermally derived SKPs labeled with CM-DiI were injected into the animal model through the femoral vein of rats. After being implanted, the SKPs were killed on the 1st, 3rd and 5th day, respectively. The frozen sections were made. Immunofluorescent staining was used to detect the expression of SKPs in the brain Survival, migration and distribution in ischemic rats. Results In the frozen section of brain tissue of MCAO rats after transplanted SKPs, the transplanted SKPs were observed under the fluorescence microscope. Vein-transplanted cells are mainly distributed around the ischemic foci, occasionally transplanted cells in the contralateral brain tissue. Conclusion The SKPs transplanted via the femoral vein can survive in the ischemic brain tissue and migrate to the injured site.
